Human IL-4
Interleukin-4 (IL-4) is a critical cytokine involved in humoral immunity and allergic inflammation. It is primarily secreted by T helper 2 (Th2) cells, as well as mast cells and basophils, and plays a key role in promoting Th2 differentiation, B cell proliferation, and immunoglobulin class switching to IgE and IgG. IL-4 exerts its effects by binding to its receptor complex (IL-4Rα paired with either the common gamma chain or IL-13Rα1), activating the JAK-STAT signaling pathway—particularly STAT6—to regulate gene expression and immune responses. Additionally, IL-4 suppresses Th1 responses, modulates inflammation, and supports tissue repair. When combined with GM-CSF, IL-4 drives monocyte differentiation into dendritic cells (moDCs), particularly in vitro.
Recombinant human IL-4 is widely used in vitro to investigate Th2-driven immunity, cytokine signaling, and gene regulation. It is a valuable tool in studying allergic diseases such as asthma and atopic dermatitis and is instrumental in drug discovery efforts targeting IL-4 pathways for immune modulation.
